My hunch is that SP1 the transcription factor is unlikely to be the clue here. Yes, it's an important protein, but it's involved in keeping a very large number of genes on, so that there's not a specific disease connection.
It seems to me that Abby clearly was underlining the "heat" clue, making it sound like the invisible ink message was part of the original letter, rather than it being added later by the reader.
